On OSX, Ardour4 supports Audio Unit, LV2 and LADSPA Plugins.
The last two are automatically indexed, Audio Units need to be scanned
for explicitly.

When you first start Ardour4.2 the setup-dialog offers to scan for
plugins. Later you can do so via  Edit > Preferences > Plugins
