Ardour4 only uses LXVST_PATH (linux VST, .so) as initial default (if it’s set at first start).  After that  you need to use 
Preferences > Plugins where you can configure the path as you wish.

You also need to explicitly scan for plugins there.

For windows VST (.dll) the environment variable is VST_PATH  but that’s only for builds with windows-vst support. The same rules apply.  Just use the Preference pane.

NI Reaktor? on GNU/Linux?

If so I hazard a guess that it crashes in wine during scan,  and ends up on the blacklist. (see Window > Log)
