Yes, I can do that on Ardour ardour 2.0.2 (built from revision 1810).
It's just Ardour's standard layer features.  If you right-click in the
track within the punch area, the top part of the menu is a list of all
the regions under the mouse.  If you select any of them, you get a
submenu which among other things provides these actions: "Raise to Top
Layer", "Lower to Bottom Layer", or (if you just want to hear the take
alone without moving layers) "Audition".
